The current discourse around transgenerational epigenetics often collapses the distinction between intergenerational and transgenerational effects. When we discuss the inheritance of stress, we must account for the two massive waves of epigenetic reprogramming (the erasure of DNA methylation marks) that occur during gametogenesis and early embryonic development. For a trait to be truly transgenerational, it must persist into the F3 generation, effectively bypassing these erasure events. Much of the popular literature relies on rodent models where environmental variables are tightly controlled, yet these results rarely translate cleanly to human cohorts. In humans, the signal is frequently drowned out by genetic variance or behavioral transmission, which is essentially social learning rather than molecular inheritance. We are seeing a tension here between the desire for a biological mechanism for trauma and the strict statistical requirements of the replication crisis. Where do you stand on the validity of these markers? The F3 threshold exists because a pregnant female exposes both the F1 fetus and the F2 germ cells to the initial trigger. Only the F3 generation is entirely removed from the original environmental stimulus.
